Optimizing the metric in sensorless adaptive optical microscopy

When observing biological tissues in-depth, one has to fight, among other things, with the consequences of optical aberrations introduced by the inhomogeneities of refractive indices. In microscopy, the deformation modes of the wave front are often corrected, one after the other. In this article, we compare the performance of two metrics, i.e. of two types of fluorescence signal measurements, adapted to adaptive optics for fluorescence correlation spectroscopy.

In addition to the total fluorescence count rate, FCS gives access to the average number of molecules in the observation volume, which makes it possible to calculate the signal per molecule, or brightness (analogous to the signal of a star guide used in astronomy). The Signal / Noise measurements carried out with test solutions show that, contrarily to the count rate, the brightness is better adapted to low concentration and moderate amount of aberrations.
